Because amantadine's elimination is highly dependent on renal function, the elimination t1/2increases up to twofold in the elderly and even more with renal impairment. Dose adjustment is advised even with mildly decreased renal function. In contrast, rimantadine is metabolized extensively by hydroxylation, conjugation, and glucuronidation; 60—90% is excreted in urine as metabolites.
