Chromatographic methods include thin-layer chromatography (TLC), highperformance liquid chromatography (HPLC), and affinity chromatography. Large-scale screening is the primary advantage of TLC; other advantages of TLC are its simple instrumentation, low cost, and speed. HPLC is the most commonly used chromatographic technique for global methylation analysis. It is sensitive, quantitative, and highly reproducible and it can use fluorescent detection, but separations are slow. Affinity chromatography is an important component of ICEAMP,82 a method that allows identification of methylation changes without the necessity of knowing the target sequence region and has no need for modification-sensitive restriction enzymes.
