Calculation of Partition Coefficients

Like in the other chromatographic techniques, the capacity factor (k') determined in CPC is related to the retention volume of the solute (VR) and the mobile phase volume (Vm) by the equation:

When using the aqueous solution as eluent and knowing the volume ratio of the mobile and stationary phase, the partition coefficient is calculated as:

where Vt is the total volume of the column. Eq. (2) can be rearranged to give Eq. (3):

where tR is the retention time of the solute, ta the dead time or retention time of non-retained solutes and U the flow rate of the mobile phase. Similarly, when using the organic phase as the mobile phase, partition coefficients are then calculated as:
