One study has reported that local infusions of MK212 into the mPFC attenuates both the locomotor stimulant, and the discriminative stimulus effects, of cocaine (Filip and Cunningham 2003). Blockade of 5-HT2C receptors with RS102221 enhanced both of these effects. Recent data suggests that both effects appear to be independent of any modulation of cocaine's effects on DA function (Leggio et al. 2009). Whether 5-HT2C receptors in the mPFC play a role in mediating effects of 5-HT2C receptor ligands on cocaine or other drug self-administration or reinstatement remains to be determined.
