Propofol is an injectable sedative-hypnotic used for the induction and maintenance of anesthesia or sedation. Propofol is only slightly soluble in water with an octanol/water partition coefficient of 6,761:1; thus, it is formulated as an oil-in-water emulsion. The fat component of the emulsion consists of soybean oil, glycerol, and egg lecithin. The pKa of the propanol hydroxyl is 11 and the injectable emulsion has a pH of 7 to 8.5. Formulations contain either disodium eth-ylenediaminetetraacetic acid (EDTA) (0.005%) or sodium metabisulfite to retard the growth of microorganisms. EDTA is a metal chelator and patients on propofol containing EDTA for extended periods of time excrete more zinc and iron in their urine.38 The clinical consequence of this is not known but the manufacturer recommends that a drug holiday or zinc supplementation be considered after 5 days of ther-apy.39 Generic formulations of propofol may contain sodium metabisulfite as the antimicrobial agent, and patients allergic to sulfites, especially asthmatic patients, should avoid this formulation.40 Aseptic technique must be followed and unused portions of the drug must be discarded according to the
